TLC 40™ TLC 42™
TLC 40™ – Cuvette holder for absorbance spectrophotometers
The TLC 40 is a single temperature-controlled cuvette holder that will fit in any Cary spectrophotometer.

For the customer who needs something special: Independent temperature control for sample and reference

The TLC 42 product permits independent temperature control for the sample and reference cuvettes. Measure absorbance as a function of temperature, keeping the reference at the same temperature as the sample, or measure the differential effect of temperature on a sample by measuring the difference in absorption when the two cuvettes are held at different temperatures.

Higher temperatures up to 150 °C

Add /E to the model numbers of the TLC 40, TLC 42 or the TLC 50 to get a modified cuvette holder that will control temperature up to 150 °C. This option requires changes in the firmware of the temperature controller, modifications in magnetic stirring mechanism to move the motor away from excess heat and changes in certain internal components to make them tolerate higher temperatures.

“ z” height variants for special cuvettes
The temperature-controlled cuvette holders for the Varian product line have a standard “z” height of 8.5 mm between the bottom of the cuvette and the optical center line of the spectrometer. This minimizes the volume required in a standard one cm square cuvette and matches a standard z height for microcuvettes. Alternatively, for a small additional cost, the TLC 40, TLC 42, TLC 50 and TURRET 400 may be also made to a z height of 15 mm, another standard height for microcuvettes. The single cell TLC 40, TLC 42 and TLC 50 can also be made for a z height of 20 mm to match cuvettes sold by Varian. If you want one of these special z height cuvette holders, add a /Z15 to the model number to get a 15 mm z height or /Z20 for a 20 mm z height.

One cuvette holder for either the Cary 50 or the Eclipse
We now offer an economical solution to the customer who wishes to purchase a single cuvette holder and use it for both absorbance and fluorescence measurements. A TLC 50/Univ is a cuvette holder that may be used in both the Cary 50 UV/Vis spectrophotometer and the Eclipse Fluorometer. Purchase this powerful pair of spectrometers with uncompromising temperature control and add another cuvette holder later when your budget permits.

4-windowed cuvette holder for a UV/Vis spectrophotometer
For certain experiments, one may wish to use a cuvette holder with additional windows perpendicular to the light beam of a UV/Vis spectrophotometer. Perhaps there is a need to pass a laser beam through the sample or perhaps to measure light emissions from the sample while an absorbance measurement is being made. Since the 4-windowed TLC 50 has the same outer dimensions as the TLC 40, it may be used in its place. The TLC 50 has four optical ports 12 mm high by 10 mm wide, one on each side of the cuvette. It also has optical slits to limit the extent of light entering or leaving the cuvette. Contact us if you would like to replace any cuvette holder of a TLC 40 or TLC 42 product with a 4-windowed version.

Low temperature operation
Operating inside of a Varian spectrometer with a flow of dry gas on the windows and circulating ice water to remove heat from the Peltier unit, these cuvette holders will operate down to about -25 °C. For frequent work at any temperatures below about -10 °C or so with the TLC 40, TLC 42 or the TLC 50 single cuvette holders, we recommend the use of a windowed jacket to isolate the cuvette. A windowed jacket is a block of insulating plastic with AR-coated fused silica windows to permit light to enter or leave the cuvette. Use a WJ-40 for a TLC 40 or TLC 42, or a WJ-50 for a TLC 50. With a windowed jacket and pre-cooled circulating fluid at a temperature of about -15° these cuvette holders will function well down to -40 °C. Even lower temperatures may be used. Contact us for advice on how to work at very low temperatures.

Items included with each purchase of a Cuvette holder for a Varian Spectrometer:

Temperature Controller
The price of each temperature-controlled cuvette holder includes a temperature controller. A single cell TLC 40 or TLC 50 is provided with the TC 125 single temperature controller. A dual-cell TLC 42 is provided with the TLC 201 dual controller providing completely independent temperature control of the sample and reference cuvettes in a dual-beam spectrophotometer. A TURRET 400 is provided with the TC 425, which combines the functions of the TC 125 with four position buttons on the front panel that may be used to rotate different cuvettes into the beam.

An initial instrumental gain calibration matches an individual cuvette holder with its specific temperature controller. Temperature performance is then tested at -20, 0, 35, 65 and 100 °C using a NIST-traceable thermometer inserted into an ethylene glycol-water solution in a cuvette placed in the holder. Comparisons between the controller read out and the NIST-traceable thermometer provide a measure of temperature accuracy. The standard deviation of the individual temperature readings returned by the controller at each calibration temperature provides a measure of precision. Typical reported values are on the order of ±0.005 °C, exceeding the ±0.02 °C performance listed in the specifications.

Temperature probe capability

The TC 125 and TC 425 temperature controllers have provision for use of an external temperature probe. Standard Series 400 and 500 thermistor probes are available from Yellow Springs Instrument Company or Cole Parmer Instrument Company. When the plug of the standard probe is inserted in the back of the temperature controller, the controller senses the presence of the probe and displays the probe reading on the LCD readout on the front panel. A variety of probes are available, such as the number 423 cuvette probe or the very small diameter 500 series probes that can be inserted into a small volume cell.

Naturally, on changing the temperature, the sample temperature lags behind the temperature of the cuvette holder. Use of a probe verifies that the desired temperature has been reached before a measurement is made. A probe also permits accurate temperature ramping experiments, because changing spectroscopic properties can be plotted vs the actual sample temperature instead of the temperature of the holder, as is done in many spectrometers.

BATH 100

Every temperature-controlled cuvette holder for a Varian instrument shipped to the continental US or Canada is provided with a BATH 100 to circulate water through the Peltier unit. The BATH 100 consists of a submersible pump, a plastic bucket and the necessary fittings. Connect tubing from the pump to the sample holder, put the pump in the bucket, and add water in the bucket. As necessary, add ice to the bucket to keep the circulating water sufficiently cool. Ice water in the bucket provides sufficient heat extraction from the Peltier to permit work at about -25 °C. At higher temperatures the bath removes heat from the Peltier unit any time the temperature is lowered.

SER 2.3 Serial Control

A Serial Control option is included with each shipment. The Serial Control Program V2.2 permits optional computer control. The program will plot temperatures against time and run scripts of simple commands to operate the controller or perform more complex functions such as temperature ramping. The serial option includes an interface within the temperature controller, a cable to connect the controller with a computer and a windows-based program for delivering serial commands to the controller. The program runs independent of the Varian software. A full range of serial commands are accepted.
