Items included with each purchase of a cuvette holder for a PTI fluorometer:

Temperature Controller
The price of each temperature-controlled cuvette holder includes a temperature controller. A single cell TLC 50/PTI is provided with the TC 125 single temperature controller. A TURRET 400/PTI is provided with the TC 425, which combines the functions of the TC 125 with four position buttons on the front panel that may be used to rotate different cuvettes into the beam.

An initial instrumental gain calibration matches an individual cuvette holder with its specific temperature controller. Temperature performance is then tested at 20, 0, 20, 35, 50 and 100 ºC using a NIST-traceable thermometer inserted into an ethylene glycol-water solution in a cuvette placed in the holder. Comparisons between the controller read out and the NIST-traceable thermometer provide a measure of temperature accuracy. The standard deviation of the individual temperature readings returned by the controller at each calibration temperature provides a measure of precision. Typical reported values are on the order of ±0.005 °C, exceeding the ±0.02 °C performance listed in the specifications.

Temperature probe capability
The TC 125 and TC 425 temperature controllers have provision for use of an external temperature probe. Standard Series 400 and 500 thermistor probes are available from a number of sources. When the plug of the standard probe is inserted in the back of the temperature controller, the controller senses the presence of the probe and displays the probe reading on the LCD readout on the front panel.

On changing the temperature, the sample temperature lags behind the temperature of the cuvette holder. Use of a probe verifies that the desired temperature has been reached before a measurement is made. A probe also permits accurate temperature ramping experiments, because changing spectroscopic properties can be plotted vs the actual sample temperature instead of the temperature of the holder, as is done in many spectrometers.

SER 2.3 Serial Control
A Serial Control option is included with each shipment. The Serial Control Program V2.2 permits optional computer control. The program will plot temperatures against time and run scripts of simple commands to operate the controller or perform more complex functions such as temperature ramping. The serial option includes an interface within the temperature controller, a cable to connect the controller with a computer and a windows-based program for delivering serial commands to the controller. The program runs independent of the PTI software. A full range of serial commands are accepted.

For the customer who needs something special:

Higher temperatures up to 150 °C
Add /E to the model number, making it TLC 50/PTI/E to get a modified cuvette holder that will control temperature up to 150 °C. This option requires changes in the firmware of the temperature controller, modifications in magnetic stirring mechanism to move the motor away from excess heat and changes in certain internal components to make them tolerate higher temperatures.

“ z” height variants for special cuvettes
The temperature-controlled cuvette holders for the PTI product line have a standard “z” height of 8.5 mm between the bottom of the cuvette and the optical center line of the spectrometer. This minimizes the volume required in a standard one cm square cuvette and matches a standard z height for microcuvettes. Alternatively, for an additional $150, the TLC 50 and TURRET 400 may be also made to a z height of 15 mm, another standard height for microcuvettes. When ordering, add a /Z15 to get TLC 50/PTI/Z15 or TURRET 400/PTI/Z15.

Low temperature operation

Operating inside of a PTI fluorometer with a flow of dry gas on the windows and circulating ice water to remove heat from the Peltier unit, either the single cell or turret will operate down to about -25 °C. For frequent work at any temperatures below about -10 °C or so with the TLC 50, we recommend the use of a WJ-50 windowed jacket to isolate the cuvette. A windowed jacket is a block of insulating plastic with four AR-coated fused silica windows to permit light to enter or leave the cuvette. With a windowed jacket and pre-cooled circulating fluid at a temperature of about -15° these cuvette holders will function well down to -40 °C. Even lower temperatures may be used.
